چکیده

All reagents were purchased from Sigma-Aldrich or Life Technologies and were used as received. Where described below pet. refers to petroleum ether (40–60 °C). All reactions were monitored by thin-layer chromatography (TLC) on pre-coated SIL G/UV254 silica gel plates (254 m) purchased from VWR. Flash column chromatography was carried out with Kiesegel 60 M, 0.04/0.063 mm (200–400 mesh) silica gel. 1 H and 13 C NMR spectra were recorded at ambient temperature on a Bruker Avance 600 instrument operating at a frequency of 600 MHz for 1 H and 150 MHz for 13 C in CDCl3. The chemical shifts () for 1 H and 13 C are quoted relative to residual signals of the solvent on the ppm scale. 1 H NMR peaks are reported as singlet (s), doublet (d), triplet (t), quartet (q), quintet (quint.), sextet (sext.), octet (oct.), multiplet (m), broad (br), doublet of doublets (dd), doublet of triplets (dt), AB quartet (ABq). Coupling constants (J values) are reported in Hertz (Hz) and are H-H coupling constants unless otherwise stated. Signal multiplicities in 13 C NMR were determined using the distortionless enhancement by phase transfer (DEPT) spectral editing technique. Infrared spectra were obtained on a Perkin Elmer Spectrum 100 FTIR Spectrometer operating in ATR mode with frequencies given in reciprocal centimetres (cm -1 ). Melting points were measured with a Gallenkamp apparatus and are uncorrected. Mass spectra were obtained on a VG70-SE mass spectrometer. Recombinant native sequence albumin 1 was obtained from Novozymes Biopharma.

Reversible protein affinity-labelling using bromomaleimide-based reagents† †Electronic supplementary information (ESI) available: 1H and 13C spectra for all new and known compounds, and ES-MS spectra for all reactions with proteins described herein. See DOI: 10.1039/c3ob40239hClick here for additional data file.

Reversible protein biotinylation is readily affected via conjugation with a bromomaleimide-based reagent followed by reductive cleavage. The intermediate biotinylated protein constructs are stable at physiological temperature and pH 8.0. Quantitative reversibility is elegantly delivered under mild conditions of using a stoichiometric amount of a bis-thiol, thus providing an approach that will b...

Acid-cleavable thiomaleamic acid linker for homogeneous antibody–drug conjugation† †Electronic supplementary information (ESI) available: 1H and 13C spectra for all novel compounds, and ES-MS spectra for all reactions with proteins described herein. In this communication we describe a novel acid-cleavable linker strategy for antibody-drug conjugation. Functional disulfide bridging of the single interchain disulfide bond of a trastuzumab Fab fragment yields a homogeneous antibody-drug conjugate bearing a thiomaleamic acid linker. This linker is stable at physiological pH and temperature, but quantitatively cleaves at lysosomal pH to release...

A novel approach to the site-selective dual labelling of a protein via chemoselective cysteine modification† †Electronic supplementary information (ESI) available: LC-MS, ES-MS, deconvoluted spectra and fluorescence emission spectra for all reactions with proteins described herein. Fluorescence emission spectra of superfolder GFP, all the cysteine mutants and their derivatives are given. See DOI: 10.1039/c3sc51333e Click here for additional data file.

Local protein microenvironment is used to control the outcome of reaction between cysteine residues and 2,5-dibromohexanediamide. The differential reactivity is exploited to introduce two orthogonal reactive handles onto the surface of a double cysteine mutant of superfolder green fluorescent protein in a regioselective manner. Subsequent elaboration with commonly used thiol and alkyne containi...

A rapid, site-selective and efficient route to the dual modification of DARPins† †Electronic supplementary information (ESI) available: LC-MS spectra for all reactions with proteins described herein. See DOI: 10.1039/c4cc00053f Click here for additional data file.

Designed ankyrin repeat proteins (DARPins) are valuable tools in both biochemistry and medicine. Herein we describe a rapid, simple method for the dual modification of DARPins by introduction of cysteine mutations at specific positions that results in a vast difference in their thiol nucleophilicity, allowing for clean sequential modification.
